Материалом для изучения послужили 27 образцов туберкулем легкого. В зависимости от степени воспалительных и репаративных процессов, происходящих в ткани туберкулемы, образцы были разделены на 3 группы. Визуализация таргетных белков проводилась с помощью иммуногистохимического метода исследования. Для количественной оценки проводился подсчет относительной площади экспрессии сигнальных молекул. Статистическая обработка полученных данных проводилась с использованием непараметрических критериев.</p></sec><sec><title>Результаты</title><p>Результаты. Анализ полученных данных показал повышение уровня экспрессии CaD при эпителизации стенки по сравнению с группой отсутствия соединительнотканной репарации, что связано с участием CaD в процессах, сопутствующих репарации и фиброзированию. При воспалительном процессе в гранулеме обнаружены статистически значимые различия уровней экспрессии интегрина αV между группами 2 и 3. Снижение уровня белка от 2 к 3 группе может быть связано с активацией противовоспалительного медиатора TGFβ1 и индукцией макрофагов М2. Результаты исследования уровня экспрессии TIMP-1 продемонстрировали его участие как в фиброзировании гранулем, так и в формировании соединительнотканного комплекса в очаге воспаления. Полученные данные свидетельствуют, что изученные белки могут рассматриваться в качестве информативных биомаркеров в молекулярной диагностике и оценке прогноза развития туберкулезного процесса в легких.</p></sec></abstract><trans-abstract xml:lang="en"><sec><title>The objective</title><p>The objective: to study the role of CaD proteins, αV integrin and TIMP-1 in the development of pulmonary tuberculosis.</p></sec><sec><title>Subjects and Methods</title><p>Subjects and Methods. 27 specimens of pulmonary tuberculomas were used for this study. The specimens were divided into 3 goups in accordance with the degree of inﬂammatory and reparative changes occurring in the tuberculoma tissue. The targeting proteins were visualized by immunohistochemical methods. For quantiﬁcation, the relative area of expression of signaling molecules was counted. The obtained data were statistically processed based on nonparametric criteria.</p></sec><sec><title>Results</title><p>Results. Data analysis showed an increased level of CaD expression during wall epithelialisation compared to the group with no connective tissue repair, which was associated with the involvement of CaD in the concomitant repair and ﬁbrosis formation. Statistically signiﬁcant diﬀerences in the expression levels of αV integrin between Groups 2 and 3 were found in the case of granuloma inﬂammation. The decrease in the protein levels from Group 2 to Group 3 may be due to activation of the anti-inﬂammatory mediator TGFβ1 and induction of M2 macrophages. The results of testing TIMP-1 expression level demonstrated its involvement both in granuloma ﬁbrosis and formation of connective tissue complex in the inﬂammation site.
